Aldosterone synthase gene C-344T polymorphism and the risk of development and chronic heart failure flow

Background. Despite considerable advances in the treatment of CAD and CHF, the prognosis for these diseases remains extremely serious, therefore the timely prevention and early diagnosis are still relevant. Objective. To study the clinical and genetic aspects of aldosterone synthase gene C-344T polymorphism and its effect on the risk of development and chronic heart failure flow. Materials and methods. The study included 226 patients with CHF (149 men and 77 women, mean age 55.9±5.8 years). Identification of genotypes was performed using RFLP analysis of PCR products. The control group included 136 people (63 men and 73 women, average age 53.6±4.8 years) who had no symptoms according to a survey of cardiovascular disorders. Results. The study revealed that patients compared with control group had significantly higher prevalence of genotype T / T of polymorphic locus C-344T of aldosterone synthase gene and the rate of C / T genotype was significantly lower. Thus, T / T genotype is associated with symptoms of CHF, and genotype C / T behaves as a protective factor. In addition, the T allele and T / T genotype are associated with the severity of clinical manifestations of heart failure. Also C / T genotype and T allele are the factors of unfavorable CHF flow, while C / C genotype and C allele are associated with favorable flow. The results of this study convincingly demonstrated that aldosterone synthase gene polymorphism in patients with chronic heart failure is associated with high individual risk of CHF development and type of this disease flow.
